What is NAD+ Optimization Strategies?

NAD+ levels decline by roughly 50% by middle age, driven by decreased NAMPT expression, increased CD38 activity, accumulated DNA damage consuming NAD+ via PARP, and decreased NRK activity. This decline impairs mitochondrial function, DNA repair, and sirtuin activity. Optimization strategies include precursor supplementation (NMN or NR), exercise, and lifestyle modifications. Human trials show NAD+ levels can be boosted, but whether this translates to meaningful health outcomes remains inconsistent.

Is there evidence for it?

EVIDENCEmoderate

How do you do NAD+ Optimization Strategies?

  1. 1.Choose a precursor: NMN (250-1000mg/day, Sinclair's choice) or NR (250-500mg/day, ChromaDex Tru Niagen); NMN may be slightly more direct as it is one step closer to NAD+ in the biosynthesis pathway
  2. 2.Support NAD+ synthesis naturally: aerobic and resistance training upregulate NAMPT (the rate-limiting enzyme), raising skeletal-muscle NAD+ by up to 25-30%; exercise is the strongest non-supplement NAD+ booster
  3. 3.Reduce NAD+ consumption: manage chronic inflammation (CD38 is the primary NADase upregulated by inflammatory signals); limit alcohol (depletes NAD+ through metabolism); optimize sleep (NAD+ follows circadian rhythms)
  4. 4.Stack synergistically: combine NMN/NR with resveratrol (sirtuin activator) and TMG/betaine (methyl donor to offset methylation burden from NAD+ metabolism); take precursors in the morning to align with circadian NAD+ peaks
  5. 5.Test and track: measure intracellular NAD+ levels (Jinfiniti test) at baseline and after 3 months to verify your protocol is working — individual response varies significantly

How long does it take?

5 minutes daily (supplements) + regular exercise.

Is it hard to start?
