Rapamycin for Longevity (mTOR Inhibition)

What is Rapamycin for Longevity (mTOR Inhibition)?

Rapamycin (sirolimus) is the only drug proven to extend lifespan in every organism tested, from yeast to mice. It works by inhibiting mTORC1, a nutrient-sensing pathway that drives growth but also accelerates aging when chronically activated. The longevity protocol uses low-dose, intermittent (weekly) dosing to selectively inhibit mTORC1 while minimizing mTORC2 suppression (which causes insulin resistance). Clinical evidence in healthy humans remains unestablished — this is genuinely experimental.

How do you do Rapamycin for Longevity (mTOR Inhibition)?

  1. 1.Work with a longevity-focused physician (mandatory) — rapamycin is a prescription immunosuppressant; self-dosing is reckless and potentially dangerous
  2. 2.Typical longevity protocol: start at 2-3mg once weekly, titrate cautiously to 5-6mg weekly based on bloodwork; some protocols use 5-10mg biweekly instead
  3. 3.Monitor labs every 3-4 months: complete blood count, comprehensive metabolic panel, lipid panel (rapamycin raises LDL and triglycerides), fasting glucose, HbA1c, liver enzymes
  4. 4.Avoid combining with grapefruit or CYP3A4 inhibitors (dramatically increases blood levels); time doses away from vaccines (may blunt immune response); do not use during active infections
  5. 5.Understand the risk profile: potential side effects include mouth sores (most common), elevated lipids, impaired wound healing, glucose dysregulation; benefit remains theoretical in humans despite strong animal data

How long does it take?

Minutes per week (dosing) + quarterly lab work.

Is it hard to start?

hard.
